Frontiers of Agricultural Science and Engineering 2015, Volume 2, Issue 3,   Pages 223-229 doi: 10.15302/J-FASE-2015065

Abstract: In recent years, the mobile metallo-β-lactamase (MBL) genes have been found to correspond to one of the most important resistance characters identified in Gram-negative bacteria, severely affecting clinical chemotherapy and threatening public health. The prevalence of mobile MBL genes and their flanking regions in Gram-negative bacteria from diseased pigs in China was investigated. A total of 334 lung samples from diseased pigs were screened for Gram-negative bacteria classified as non-susceptible to meropenem (MIC≥4 mg·L ). Six isolates, including three , two and one , exhibited MBL production and carried the gene. S1-PFGE and Southern blot analysis showed that the gene was located on the chromosome of one isolate and on plasmids of various sizes in the other five isolates. MIC testing using broth microdilution revealed that all -carrying isolates and some of their transconjugants exhibited resistance to almost all β-lactams tested. Whole genome sequencing revealed that the flanking region of the gene from all porcine isolates had high levels of similarity with the corresponding regions in human isolates. One porcine isolate carrying was typed as ST48, a common sequence type in human isolates. These results suggest the possibility of human-to-food animal transfer of -producing , highlighting the need for surveillance of carbapenemase producers among bacteria from food animals. In addition, the prudent use of antimicrobial agents to decrease the opportunities for co-selection of carbapenemase genes in food animals is also urgently needed.

lncR-GAS5 upregulates the splicing factor to impair endothelial autophagy, leading to atherogenesis

Frontiers of Medicine 2023, Volume 17, Issue 2,   Pages 317-329 doi: 10.1007/s11684-022-0931-4

Abstract: Here, we investigated the role of the lncRNA growth arrest-specific 5 (lncR-GAS5) in atherogenesis. Moreover, serine/arginine-rich splicing factor 10 (<i>SRSF10i>) knockdown increased the LC3II/LC3IMechanistically, lncR-GAS5 regulated the downstream splicing factor <i>SRSF10i> to impair autophagyIn conclusion, lncR-GAS5 overexpression arrested endothelial autophagy through the miR-193-5p/<i>SRSF10Thus, miR-193-5p/<i>SRSF10i> may serve as a novel treatment target for atherosclerosis.

5′-tiRNA-Gln inhibits hepatocellular carcinoma progression by repressing translation through the interactionprogression by repressing translation through the interaction with eukaryotic initiation factor 4A-I

Frontiers of Medicine 2023, Volume 17, Issue 3,   Pages 476-492 doi: 10.1007/s11684-022-0966-6

Abstract: In HCC cells, 5′-tiRNA-Gln overexpression impaired the proliferation, migration, and invasion <i>in vitroi> and <i>in vivoi>, while 5′-tiRNA-Gln knockdown yielded opposite results. 5′-tiRNA-Gln exertedits function by binding eukaryotic initiation factor 4A-I (EIF4A1), which unwinds complex RNA secondaryG-quadruplex structure is crucial for 5′-tiRNA-Gln to strongly bind EIF4A1 and repress translation.Clinically, 5′-tiRNA-Gln expression level is negatively correlated with ARAF, MEK1/2, and STAT3 in HCC
